A few that don't seem to have been listed yet:

Johnny Wander - a very low-key comic about two people and incidents from their real day-to-day lives that often prove amusing. Updates Tuesday and Thursday (though not always very early in the day).

Girls With Slingshots - comic about a gal who, let's face it, has something of a drinking problem, her friends, various events. Sometimes funny, sometimes more serious. Not sure of the update pattern, but it's fairly regular.

Dresden Codak - I'd list this as a contender for "Best Webcomic on the Internet." Great art, good (if sometimes difficult-to-follow) writing, fair mix of high concept and simpler comedy. Downsides: updates very slowly, starts out incredibly different from what it progresses into (compare the first page, The Tomorrow Man with Lantern Season four years later). I highly recommend you give this one a try with a little patience for the early stuff if you start at the beginning; it gets less hard-science-jokes later on.

Lackadaisycats (or, technically, just "Lackadaisy") - Seriously, how is this not listed already? Anthropomorphized cats running a speakeasy during the prohibition, all of it done in sepiatone with some of the best artwork you'll find in webcomics, particularly with regard to setting and clothing detail. Just look at the first panel on this page. Look at it. @.@
